Modulation of cGMP in retina
The focus of this research is cGMP in the inner retina. Because cGMP levels
are regulated by several distinct biochemical mechanisms, many questions
remain about which enzymatic pathways can modulate cGMP in particular
retinal neurons and the synaptic mechanisms underlying this modulation. We
are determining which specific retinal cell types contain particular enzymes
involved in cGMP mechanisms, and examining the effects of the selective
activation or inhibition of these enzymes on cGMP levels in identified
retinal cells. This will done using antisera directed against cGMP itself,
soluble guanyl cyclase subunits, isoforms of particulate guanyl cyclases,
isoforms of heme oxygenase and isoforms of nitric oxide synthase to localize
these antigens within specific cells. cGMP immunocyto-chemistry and
immuno-assays will be used to measure changes in cGMP levels in response to
physiological stimuli, or to the inhibition or activation of specific cGMP
related enzymes. We are also studying whether ganglion cell activity or
activation of efferents to the retina can modulate levels of cGMP in retinal
neurons. Finally, we are analyzing the synaptic connectivity of specific
retinal neurons associated with particular enzymatic pathways that modulate
cGMP in retina.
Candidates should have some prior training in retinal anatomy or
electrophysiology, but candidates with skills in retinal pharmacology or
biochemistry will also be considered. Qualified candidates should contact me
